"Nani?" is the equivalent of "What?" In English.  Most Americans are only aware of the word due to the "Omae Wau Mou, Shinderu." Meme.  The meaning of which is "You are already dead."  The other person responds "Nani?" Which means "What?"  Get some class and watch a subtitled anime instead of a dubbed over one.
